  • The microenvironment of Kras;D/+ mice supports myeloproliferative neoplasm development of wild type cells in a cell non‐autonomous manner. Lethally irradiated control, KrasLSL G12D/+; Mx1‐Cre (Kras) or KrasLSL G12D/+; Rosa26LSL Dnmaml‐GFP/+; Mx1‐Cre (Kras; D/+) mice (CD45.2+) were transplanted with 5 × 106 CD45.1+ wild type bone marrow cells. Moribund recipients were sacrificed for analysis. (A) Kaplan‐Meier survival curves of different groups of recipient mice were plotted against days after transplantation. P values were determined using the Log‐rank test. (B) Quantification of spleen weight and thymus weight. (C) Representative spleen histological haematoxylin and eosin stained sections from different groups of recipients are shown. (D) Flow cytometric analysis of donor‐derived cells in bone marrow, spleen and peripheral blood. Debris and unlysed red blood cells (low forward scatter) and dead cells (propidium iodide positive) were excluded from analysis. (E) Complete blood count analysis of peripheral blood samples collected from reciprocally transplanted mice. Haemoglobin concentration, haematocrit, and white blood cell, red blood cell and platelet counts are shown. The results are presented as mean ± SD. *P < 0·05; **P < 0·01; ***P < 0·001.
