  • In the presence of fluoride, [3H]inositol still labels a G protein and receptor‐sensitive pool of phosphatidylinositol. Cortical slices were incubated 2 h at 37°C in KH buffer with 10 μCi/ml [3H]inositol and in the presence of 10 mM Nal, then washed with fresh KH buffer and used to prepare membranes as indicated in Methods. 250 μg membrane protein were then incubated 20 min at 37°C as in legend to Fig. 2, but in the presence (upper panel, left) or absence (upper panel, right) of 2 mM ATP. Results in these upper panels, which are means±S.E. of triplicate determinations in a single experiment, represent total production of 3H‐inositol phosphates, and are expressed as a percentage of 3H‐inositol lipids in each sample (15,000 d.p.m.). Lower panel shows a typical HPLC separation of water‐soluble reaction products in the presence of ATP and under control conditions (open symbols) or upon stimulation of phospholipase C with 1 μM GTPγS plus 1 mM carbachol (filled symbols).
