  • Apical 22Na+ uptake in native gallbladders increases prior to gallstone formation. The influx of 22Na+ from the mucosal solution into the epithelium was measured in native gallbladder tissues from animals fed either nonlithogenic chow (CON) or 1·2% cholesterol‐enriched diet for varying lengths of time to induce cholesterol super‐saturated bile (PreCRYS), crystals (CRYS) and gallstones (GS) for 1 min using modified Ussing chambers with only mucosal side exposed to the Ringer's containing 140 mM Na+. The tissue was preincubated with Ringer's for 30 min before addition of radioisotope‐containing solution. The graph shows the baseline uptake of 22Na+ in CON, PreCRYS, CRYS and GS animals. Note significant increases in PreCRYS (∼55%, P < 0·05) and CRYS (∼28%, P < 0·05) groups compared to CON. Five tissues were used in each dietary group.
