- Degradation of iRBC in macrophages and DC. Macrophages (a, d) and DC (b, e) were incubated with DDAO‐labelled iRBC (a, b) or GFP‐expressing iRBC (d, e) at the schizont stage. After 1 h, cells were washed to remove nonphagocytosed iRBC. Cells were scraped at indicated time points and immediately analysed by flow cytometry. For DDAO‐labelled iRBC (a, b), gating was performed on all phagocytes and results are displayed as DDAO Fluorescence intensity over time. Control represents cells alone without iRBC. Histogram plots are representative of one of three independent experiments. For GFP‐expressing iRBC (d, e), gating was performed on cells that had phagocytosed. (c) As DC and macrophages autofluorescence overlaps with the GFP signal, we first analysed the forward (FSC)/side (SSC) scatter plot for iRBC, DC (or macrophages, not shown), and co‐cultures of DC (or macrophages) and iRBC for different incubation times (1·5 h is shown as example). Region 1 (containing iRBC and/or DC debris) is represented in blue in FL‐1/Fl‐2 plots below. Region 2 (containing DC) is represented in red. DC that have phagocytosed GFP‐expressing iRBC (Region 3) show higher levels of FL‐1 than, but the same levels of FL‐2 autofluorescence as those of, control cells. The percentage of DC in R3 was considered the percentage of phagocytosis for each time point (DC that have phagocytosed RBC or iRBC, but have not degraded them yet). (d, e) Results are displayed as percentage of phagocytosis (determined as explained in figure part c) over time. Data are displayed as mean ± standard deviation of triplicate samples. Results are representative of three independent experiments.
