- A) Double immunofluorescence with anti‐synemin (1, 3, 5) and fluorescent phalloidin (2, 4, 6) of U‐373 MG astrocytoma cells (1, 2), PPC1 (3, 4) and PC3M (4, 6) prostate carcinoma cells. Actin staining with fluorescent phalloidin reveals a leading edge (arrows) in the three cell lines. Synemin staining is present in the leading edge of U‐373 MG cells (1) but not in that of either PPC‐1 (3) or PC3M cells (5). Scale bars = 10 μm. B) Western blot of a 4–15% gel loaded with 20 μg of total proteins from U‐373 MG, PPC1, and PC3M cells; numbers at left indicate molecular mass (kDa). Synemin antibodies label two bands a‐ and β‐synemin in U‐373 MG cells (top and bottom bands, respectively) but do not recognize any protein in PPC1 and PC3M prostate carcinoma cells.
