Description:
Organization of Tn5‐based transposon vectors and their suicide delivery plasmid. The scheme represents the features common to all constructions, the whole being generally named as the pUT system. The transposase gene of Tn5 devoid of NotI sites is present nearby but outside the mobile element (transposon vector) which includes sites for unique and very unusual restriction enzymes SfiI/AvrII and NotI. This sequence is flanked by the two 19 bp I and O termini of Tn5. The SfiI/AvrII site is then used for the insertion of a variety of antibiotic and non‐antibiotic selection markers whereas the NotI site is used for cloning heterologous DNA fragments encoding desired phenotypes. These constructs are maintained in host strains that produce the R6K‐specified π protein. For mobilization, pUT plasmids are transformed into an E. coli strain with a chromosomally integrated RP4 that provides conjugal transfer functions which act on the oriT sequence present in the plasmid.
